HPLC vs. LC-MS in Peptide Analysis

HPLC and liquid chromatography–mass spectrometry answer different analytical questions. Neither acronym should be treated as a quality guarantee without reviewing the actual method and report.

HPLC separation

High-performance liquid chromatography separates sample components according to their interactions with a stationary phase and mobile phase. The chromatogram displays detector response over time. When the method is appropriate, peak integration can support a purity assessment, but the result remains method-dependent.

Mass-spectrometric detection

Mass spectrometry measures ions by mass-to-charge ratio. Coupling chromatography with MS can help separate components and examine whether measured ions are consistent with the expected molecular mass. Instrument type, ionization, adducts, charge states, and interpretation all influence the result.

Complementary evidence

Chromatography can describe separation and relative detector response, while MS can support molecular-identity evaluation. A robust record clearly labels which question each test addresses rather than collapsing all results into a single quality claim.
